The polydimethylsiloxane( PDMS) double-layer chip is fabricated by the micro-fabrication technology
and the chip is used to construct and culture spheroids
which can be used for drug screening and post-operative individualized treatment. The chip consists of the microwell array for cell culture and a multi-channel shunt structure capable of generating concentration gradients. The results show that it can promote cell aggregation to form three-dimensional cell spheroids with a diameter of 130 μm under the dual action of microwell limitation and surface anti-adsorption. Moreover
the structure of the spheroids is stable and good activity( the survival rate of spheroids is about 80 %). It has a good application prospect in the fields of biomedicine and clinical medicine.
